Establishment a sustainable and interminable cell collection of endangered animals for genetic conservation purpose

Conservation of genetic resources means “conservation diversity to be able to adapt to the challenges that are ahead”.The genetic diversity of livestock in the country is a very precious national asset, some of which are endangered. Caspian horse, Bactrian camel and Markhoz goat are highly endangered among the native animals. The Caspian is a small horse breed. Iranian Bactrian camel (Two Hump camel) lives in the -30 to 30° C temperature range. Markhoz goat produces the lustrous fibre known as mohair. In this project, 54, 56, 58 cell lines were generated from Caspian horse, Bactrian camel and Markhoz goat, respectively. Primary cultures were prepared from tissue samples. After three passages, the cells were frozen in media containing (90%) FBS and DMSO (10%). Absence of mycoplasma contamination of cells were confirmed by direct culture methods, DNA staining and PCR method. Cells had no cross-contamination with other species and species confirmed by PCR methods. Then, vials containing the cells were transferred to a nitrogen tank. For all samples, certificate, cell photo, animal photo were prepared. This newly established cell lines will not only preserve the genetic resources at the cell level but will also provide a valuable resource for genomic, postgenomic, somatic cloning research and created an easily accessible database for researchers and students.
