Establishment cell line & gene bank of Markhoz goat for registration and conservation of Iranian animal resources

The genetic diversity of livestock in the country is a very precious national asset, some of which are endangered. Markhoz goat is native to the regions of Kurdistan and West Azerbaijan and rare breeds of goats in the world to produce the lustrous fibre known as mohair to be endangered. In this project, samples were taken from 30 males and 30 females goats with respect to effective population size and totally 58 Markhoz goat cell lines were generated. Sampling position numbed and disinfected with 70% ethanol. Ear margin tissues (about 1 cm2 in size) were sampled with an ear notcher. Primary cultures were prepared from tissue samples in the laboratory. After three passages, the cells were frozen in media containing (90%) FBS and DMSO (10%). Absence of mycoplasma contamination of cells were confirmed by direct culture methods, DNA staining and PCR method. Cells had no cross-contamination with other species and goat species confirmed by PCR methods. Then, vials containing the cells were transferred to a nitrogen tank. For all samples, certificate, cell photo, animal photo were prepared. The results indicate that the acquisition and storage of Markhoz goat fibroblast cell lines considering the effective size of the two sexes is the best way to preserve genetic resources.
